I'll just cover the basics and not explain them.
You basicly have to send a successul naval attack, followed by a land attack and a conquest attack, and then support the city under siege. You have to defend to defend this cty for 12 hours(this depends of the worlds).
The best strategy is doing timed attacks with your alliance. Only few sieges per day.

Type of Cities
Defensive style:
x OLU/MOU city; x LS city; x Mixed Defensive City; Conquest City
Offensive style:
x OLU/MOU city; x LS city; x Mixed Defensive City
x represents a number. In the offensive style, you have to be a sniper(check advanced guides).
MOU or flyers' city, you can use them to replace an OLU city, but I don't recommend to use them so often, because you're main firepower will be made by regular units. But in conquest they have a different role: break sieges, flyers pass over birremes and often find a weak DLU inside.
Attacking
Research CS and Conquest in all your OLU and Conquest cities, this not includes city of flyers(read MOU city). Your goal is to destroy all the land units of the enemy with your OLU nuke and destroy all the naval units with LS nuke.
To make your OLU army, I recommend to build at least, 50 light ships and Fast Transport Boats. In conquest, it's common to find low level walls.
First send the LS nuke, then your OLU nuke(revolt attack) and right after your conquest attack, otherwise, you can lose many OLU and your CS for a stack of birremes.
If the first phase success, then send as much support you can, but send more birremes than DLU. If the enemy couldn't break the siege, the city is yours.
Defending
Don't upgrade your walls in all your cities(unless you're a part of a special operation - read advanced).
Save your DLU and birs for a siege, unless you're mixed defensive city is going to be attacked.
If you're OLU/MOU is going to be in a siege, then send them as an attack to an inactive city and call them back in time to break the siege, the attack have to return after the CS attack.

Under a siege, ask help for your alliance. It need to attack your city and break the siege with LS or flyers.
If you spot a CS incoming, you can stack birs in your city, sinking the CS.

Supporting
Conquest is more interesting:
As I stated before, you have to save your DLU and Birs to support sieges, or the enemy will just break your siege. Yes, your OLU will be there(but you can recall) and they aren't good in defending, unless they are mixed units. OLU and LS save them to initiate sieges and to break sieges.
- With your OLU you can attack, defend yourself and break sieges in teammates.
- With your LS you can attack, defend yourself and break sieges.
- With you DLU/Birs you can defend and support sieges.
- With conquest cities, you can defend, attack and support sieges at the same time.

Basic Strategy
In conquest worlds, it's essential to be in a good alliance. Communication and activity are the key to the success. In the first months, you're not that strong, then you need the help of your allies and help them.
